Fingerprints are biometrics. You don't need to take it again unless yours were not recieved by the USCIS or it is been more than 15 months. They would issue you another notice.
EAD renewals are not much related to fingerprinting though some claim it may trigger some activities on processing.

1) If you did not apply EAD, you are not eligible for AC-21. That means you should start the whole GC process with new job from LC. Your 485 could be denied even when your 140 is approved.

Shouldn't be such a big deal. This is temporary. When you get the real card this is not valid anymore.
dbwr is right. this doesn't concern your eligiblility on EB2 or EB3. It is about the job requiring EB2 or EB3. Since your LC is on EB3, your 140 is based on that LC so is your 485. So you are stuck with EB3.
