Can a greencard holder claim unemplyment benefits? If so will it affect obtaining citizenship status later on?

Yes for the 1st Q "Can a greencard holder claim unemplyment benefits"?

No for the second Q "If so will it affect obtaining citizenship status later on"?

Unemployment benefit, as I understand it, is something the employer pay in advance from his salary in case he/she may become unemployed in the future, it is not a gift from the government solely because you are GC holder as many think and has nothing to do with being GC holder, USC, visa ...etc
The most important factors in USC application are to fulfill the residency requirement in US with GC and criminal history. ALMOST everything else is not that critical and as said, unemployment benefit as well as retirement, social security benefits for example are something you saved for your future and has nothing to do with eligibility for citizenship
 
but if you receive unemployment benefits, wouldn't the government then turn around and then charge your GC sponsors for it and make them pay? I thought that was the reason they asked for sponsors in the first place. I am very curious about whether that would be an issue.

Sponsorship is for means-tested benefits, which UI is not. It is paid for (mostly) out of employer premiums.

Sponsor your GC has nothing to do with UI. Sponsor means that you are qualified to work for your employment and benifit the US economy by your work and because of that the US give you GC and even USC after some time to work not to stay home. UI is some thing you do for your self like insurance for example in case you will become unemployed in the future.
