Maybe. You should look at the reasons you were denied and see if those have been improved. For example, if you were not a tenur-track faculty they might think you did not have a permenant position, and now you have it.
You meet the requirement of advance degree. If you are talking about EB2 NIW, you need to meet three additional criteria - your field has intrinsic value, you made contribution to the field and your leaving has adverse effect. These have to do with your work and your achievment.
I think it would be hard to do that. 485 cannot be filed when the priority date is not current. That is just my understanding. You may try infopass to see what the officer says about your status.
