My sister married a US Citizen and she is now a permanent resident.Can THEY sponsor me? What types of Visas am eligible for? I am a Japanese Citizen.

Only your sister (when she becomes a US citizen) can sponsor you, her husband can't.

Lets say beneficiary is in US on H1/H4, Will he/she is be eligible to work without H1 or apply for EAD ?
Not for 12-14 years.

Nope. He/she has to maintain the valid H-1 status in order to work legally in the U.S.
