When you apply for a derivative visa do you have to file for an affidavit of support as well, after becoming a LPR?
Does it mean that you have to live in the US when you apply for it & do a job in US also ?

In order to file a petition for a relative, yes you need to be in the US to do that. And yes an affidavit of support is required, however you don't necessary need to personally have or meet the required level of income in other to petition a relative, you can always have a co-sponsor.
Also the affidavit is not required at the time of submitting the I-130, which is the first step in petitioning for a relative. The I-130 can be filed as soon as you become a LPR, (again bear in mind that proof of income is not required at this stage) and upon it's approval you will then have to wait for a visa to become available in that category.
