my brother has a B-2 visa, I want to petition him, I want to know if he can stay in the US and get a work permit while he wait for the process.

Assuming you are a US citizen, you can indeed sponsor him for a GC. However, that family category is severely backlogged. He is looking at a 12-15 years wait. He will not be eligible for an EAD (work permit) during this period. Plus he needs to maintain lawful non-immigrant status if he decides to wait in the country. Otherwise, he has no choice but to wait outside.
