Hi,

1) For a mail-in transfer of TN status application, are you allowed to start working for the new employer as soon as you obtain the Receipt No or do you have to wait for the TN status approval and then subsequent receipt of new I-94?

2) Does the 240 day rule apply as soon as you receive the receipt no until your TN status is approved?

There is no such thing as a "TN Transfer". You are seeking employment with a new TN employer, and cannot start until the TN is approved.

If you are seeking extension of an existing TN petition with the same employer, the 240 day clock starts after the existing TN expires.
