For the new I-140 amendment you need to contact your employer
to file the new petition and refer to earlier LC, I-140 and I-485.
This will not change the priority date.
Layoff or change of job beyond 180 days after filing I-485 is not a problem.
If layoff for I-485 receipt

Hi,
Amendment I-140 will not alter I-485 infact some of my friends
who in the past applied for amendment I-140 was approved with GC
when their case date became current without waiting for I-140 to be
approved.
Technically if your 485 has to be approved in the given time frame
all they check is if there was need to file amendment I-140 and was
if filed and not wait for I-140 processing time line.

Best wishes.
