Time is very critical for me. My brother will turn 21 very soon. And, I also don't wanna get denied by because of early filing. So, when is the best time to file? Let's say my 5 years PR date is 2/22/2010. When should I start filing the citizenship application?

You can file 90 days before you meet the continuous residency requirement unless you are in the military or are filing as a PR spouse of a US citizen. You can now do the math.
