I am starting employment with a new employer on H1. My h1 transfer has
been filed under regular processing and it may take a couple of days
to get it approved. My question is can I start the Labor processing
(for green card) without the approval in hand?
 
To start your GC processing first of all you no need to be in USA.
Your future employer can process a labor certification (GC) process while you are in your home country.

An H1 is not needed to initiate the LC process. Start working on it now. You can even process the LC and I-140 before joining, if need be.
