I am USC and married to a green card holder. She has applied for her son back in 2010. Son is 27 years old. We got married 2 months back. What needs to be done to expedite the GC application of my step son?

There is no rush. Adult son petitions 2nd preference take several years. If/When she becomes a citizen she can upgrade her petition to citizen instead of permanent resident so that its 1st preference however that's not going to change much, it can even get longer. You're still looking at about six to seven years depending on country.

Good luck.

Nothing can be done to speed it up, except her getting US citizenship as soon as she can. You're not eligible to file for him, because you got married when her son was already over 18.

However, the wrong actions can slow down or destroy the case. Is he inside the US? If inside, legally or illegally? Is he planning to marry in the near future? Those factors affect the actions that need to be taken to preserve his eligibility.
