During Biometrics they will take your Thumb print and a photo.(same they do during passport stamp). They also doing biometrics with all recent fingerprinting and also some with EAD renewals.

Biometrics was earlier done during passport stamping after 485 approval. Looks like USCIS introduced a new system to eliminate passport stamping system. I got the FP+ biometrics recently with a code 3 where as my daughter got the same FP notice for biometrics only.
