When invoking AC21, Do you need to include all the dependants ? What can happen, if the dependant are not included in AC21 Letter? Is it mandatory or optional?

It's completely irrelevant. The dependents are only tied to the primary I-485 applicant; not to any job offer. They do not need to be mentioned at all.
