I've already applied for 485. But I wish to use the AC-I140 route for CP at Mumbai. Do me and my spouse have to do the medical exam all over again ? Or Can we just use the results of the 485 medical stuff ?
Nope, you would have to re-take the medical examination at Mumbai by the doctors/hospital designated by US Consulate, Mumbai.

...if you/your wife have taken certain vaccinations here in the US during the medicals, then suggest take a certificate from the doctor & carry it with you to Mumbai to avoid taking the same shots again.
