Your wife need not do anything special unless she wants to travel outside US. Because the H1 is issued when she is already in the country, if she sends her passport to DC for visa stamping, then she will get single entry stamp. So, what people usually do is go outside US (canda or your home country) and go for visa stamping there. This is just my opinion.
