My company has less than 5 employees and is there any general rule or regulation or unwritten rule that indicates the minimum number of employees of a supporting employer for employee's I-140/I485?

You are likely to get a RFE for ability to pay. This is common for small companies. My company have over 200 employees. Even then some people have got RFE on this issue.
