Can one use the already approved labor under PERM processing(as a substitute labor) and apply for I140.

maybe not, I heard only under old LC system, you can use sub., which ended 03/28/05.

so far USCIS hasn't published anything saying substitution isn't possible anymore. They have some stuff in the works and it may be forthcoming pretty soon, but as of now, it is still possible.

Wrong. I still can use Sub Labor last month.
