Dept of State - which mailing service

Does it make any difference in waiting time, which courier service I will choose for sending my documents to St Louis ?
 
I see there is a box number in the Dept of State address in St Loius for courier, non-postal deliveries. Do I then need to sign "Release signature" section on the airbill to authorize delivery without signature ? If I don't, will it cause any problems or delays ?
