applying green card for the future post

I was not having 5 years of experience when I joined my current employer, He applied green card for future post . I would like to know when the INS asks about promotion? Or they wont ask at all? Is that same as asking employement status? or it differs.

Your question is little unclear. What I could understand is, when you joined the company you did not have 5 years of experience. In your present company you completed your 5 yrs of expereince and applied labor for a future position (probably a senior position) then you should get promoted to the senior position at the time (or anytime before) you get you green card (at the time of stamping after I485 approval)
