"Are either of your parents US citizen?"

My dad is a citizen but my mother is not. Should I answer yes to this question or no.

You should answer "Yes".

When did your dad become a citizen, before or after you were born? If after, how old were you when he became a citizen and did you have a green card at the time?
Before filing N-400 you need to check whether you are a U.S. citizen already.
 
They ask those questions because if at least one of your parents was a citizen before your birth, or before you turned 18, it is possible that you already automatically derived citizenship through them. Which if true, they may deny your N-400 and tell you to establish proof for derivative citizenship. So, have you researched the rules on derivative citizenship to determine if you are already a citizen?
