As for marriage base green card , does authorised doctor checks that female spouse is virgin or not ? Second can doctor or immigrant officer require evidence for having sexual relation between husband and wife at the time of medical eximination by the doctor and at the time of final interview by the immigrant officer

1.What you describe is generally not part of the applicant's pre-interview medical exam. Without knowing your country, hard to respond accurately.
2.The interviewing officer can ask probing questions if he has doubts a bona fide marriage exists. In general, a married couple would be expected to consummate their marriage if they're physically together.
